    Unveiling the Efficiency of Vermicompost Derived from Different Biowastes on Wheat (Triticum aestivum L.) Plant Growth and Soil Health

    The present study was conducted to explore the role of different types of vermicomposts (VCs) prepared from different substrates to improve soil health (physical and chemical properties) and wheat plant growth under field conditions. Different combinations of vermicompost prepared from different substrates (cow dung, paper waste, and rice straw) and inorganic fertilizers were applied in soil using wheat as a test plant. The impact of three different VCs on physico-chemical characteristics and nutrient availability in soil was evaluated to examine their efficacy in combination with chemical fertilizers. Temporal trends in vermicomposting treatments at various stages showed significant improvement in physico-chemical attributes of the VCs substrates. All the plant physiological attributes showed significant response where N: P: K 100:50:50 kg ha?1 + 10 t ha?1 cow dung vermicompost was applied. In addition, post-harvest analysis of soil not only revealed that different combinations of the vermicomposting treatments improved the soil health by improving the physico-chemical attributes of the soil. Conclusively, application of cow dung vermicompost along with recommended NPK not only improved crop yield, soil health, reduced insect (aphid) infestation but also fortified grains with Zn and Fe. © 2019 by the authors.7527, 9017; Provozn?     Analysis of the Mutual Trade Between the Eu and Indonesia

    Mutual trade between EU countries and Indonesia is a bilateral commodity trade flow exceeding 17 billion Euros. Analysis of the trade between the two entities is an important contribution leading to the definition of the significance that the ongoing cooperation between the EU and Indonesia has. EU is for Indonesia very important trading partner, for EU is Indonesia a strategic political partner. The bilateral trade is currently greatly benefiting Indonesia, however, this market has a potential for EU due to the fact, that it is one of the most populous country in the Asian region and the in world in general. The trade exchange is based on export and import of most basic aggregate commodities; while between the two partners exist significant differences in terms of available comparative advantages. A specificity of the mutual trade is the limited territorial structure of this exchange, as the main volume of transactions is realized between Indonesia and only a few EU countries.     Inhibition of Hedgehog signaling sensitizes NSCLC cells to standard therapies through modulation of EMT-regulating miRNAs

    Abstract Background Epidermal growth factor receptor- tyrosine kinase inhibitors (EGFR-TKIs) benefit Non-small cell lung cancer (NSCLC) patients, and an EGFR-TKIi erlotinib, is approved for patients with recurrent NSCLC. However, resistance to erlotinib is a major clinical problem. Earlier we have demonstrated the role of Hedgehog (Hh) signaling in Epithelial-to-Mesenchymal transition (EMT) of NSCLC cells, leading to increased proliferation and invasion. Here, we investigated the role of Hh signaling in erlotinib resistance of TGF-β1-induced NSCLC cells that are reminiscent of EMT cells. Methods Hh signaling was inhibited by specific siRNA and by GDC-0449, a small molecule antagonist of G protein coupled receptor smoothened in the Hh pathway. Not all NSCLC patients are likely to benefit from EGFR-TKIs and, therefore, cisplatin was used to further demonstrate a role of inhibition of Hh signaling in sensitization of resistant EMT cells. Specific pre- and anti-miRNA preparations were used to study the mechanistic involvement of miRNAs in drug resistance mechanism. Results siRNA-mediated inhibition as well as pharmacological inhibition of Hh signaling abrogated resistance of NSCLC cells to erlotinib and cisplatin. It also resulted in re-sensitization of TGF-β1-induced A549 (A549M) cells as well the mesenchymal phenotypic H1299 cells to erlotinib and cisplatin treatment with concomitant up-regulation of cancer stem cell (CSC) markers (Sox2, Nanog and EpCAM) and down-regulation of miR-200 and let-7 family miRNAs. Ectopic up-regulation of miRNAs, especially miR-200b and let-7c, significantly diminished the erlotinib resistance of A549M cells. Inhibition of Hh signaling by GDC-0449 in EMT cells resulted in the attenuation of CSC markers and up-regulation of miR-200b and let-7c, leading to sensitization of EMT cells to drug treatment, thus, confirming a connection between Hh signaling, miRNAs and drug resistance. Conclusions We demonstrate that Hh pathway, through EMT-induction, leads to reduced sensitivity to EGFR-TKIs in NSCLCs. Therefore, targeting Hh pathway may lead to the reversal of EMT phenotype and improve the therapeutic efficacy of EGFR-TKIs in NSCLC patients

    The Side Population in Human Lung Cancer Cell Line NCI-H460 Is Enriched in Stem-Like Cancer Cells

    Lung cancer is among the most lethal malignancies with a high metastasis and recurrence rate. Recent studies indicate that tumors contain a subset of stem-like cancer cells that possess certain stem cell properties. Herein, we used Hoechst 33342 dye efflux assay and flow cytometry to isolate and characterize the side population (SP) cells from human lung cancer cell line NCI-H460 (H460). We show that the H460 SP cells harbor stem-like cells as they can readily form anchorage-independent floating spheres, possess great proliferative potential, and exhibit enhanced tumorigenicity. Importantly, the H460 SP cells were able to self-renew both in vitro and in vivo. Finally, we show that the H460 SP cells preferentially express ABCG2 as well as SMO, a critical mediator of the Hedgehog (HH) signaling, which seems to play an important role in H460 lung cancer cells as its blockage using Cyclopamine greatly inhibits cell-cycle progression. Collectively, our results lend further support to the existence of lung cancer stem cells and also implicate HH signaling in regulating large-cell lung cancer (stem) cells

    Epithelial to Mesenchymal Transition by TGFβ-1 Induction Increases Stemness Characteristics in Primary Non Small Cell Lung Cancer Cell Line

    Cancer Stem Cells (CSCs) hypothesis asserts that only a small subset of cells within a tumour is capable of both tumour initiation and sustainment. The Epithelial-Mesenchymal Transition (EMT) is an embryonic developmental program that is often activated during cancer invasion and metastasis. The aim of this study is to shed light on the relationship between EMT and CSCs by using LC31 lung cancer primary cell line.A549 and LC31 cell lines were treated with 2 ng/ml TGFβ-1 for 30 days, and 80 days, respectively. To evaluate EMT, morphological changes were assessed by light microscopy, immunofluorescence and cytometry for following markers: cytokeratins, e-cadherin, CD326 (epithelial markers) and CD90, and vimentin (mesenchymal markers). Moreover, RT-PCR for Slug, Twist and β-catenin genes were performed. On TGFβ-1 treated and untreated LC31 cell lines, we performed stemness tests such as pneumospheres growth and stem markers expression such as Oct4, Nanog, Sox2, c-kit and CD133. Western Blot for CD133 and tumorigenicity assays using NOD/SCID mice were performed.TGFβ-1 treated LC31 cell line lost its epithelial morphology assuming a fibroblast-like appearance. The same results were obtained for the A549 cell line (as control). Immunofluorescence and cytometry showed up-regulation of vimentin and CD90 and down-regulation of cytocheratin, e-cadherin and CD326 in TGFβ-1 treated LC31 and A549 cell lines. Slug, Twist and β-catenin m-RNA transcripts were up-regulated in TGFβ-1 treated LC31 cell line confirming EMT. This cell line showed also over-expression of Oct4, Nanog, Sox2 and CD133, all genes of stemness. In addition, in TGFβ-1 treated LC31 cell line, an increased pneumosphere-forming capacity and tumours-forming ability in NOD/SCID mice were detectable.The induction of EMT by TGFβ-1 exposure, in primary lung cancer cell line results in the acquisition of mesenchymal profile and in the expression of stem cell markers

    Abstract MAITAH, M., SMUTKA, L.: Economic analysis of milk production and consumption in the Middle Milk products are considered as the essential food commodities for humans. Milk products contain essential elements for the human body such as protein, glucose, minerals and vitamins. Moreover, milk is considered the cheapest source of animal protein, an important resource for some related transformation industries and provides employment opportunities for a large number of small producers in both rural and urban areas. The aim of this paper is to analyze the factors which determine the supply and demand for liquid milk (henceforth milk) in the Middle East and North Africa in order to point out the main problems and constraints obstructing the milk production in this region. The research also attempts to understand the drivers for the development in milk production in the Middle East and North Africa.Total milk production in the Middle East and North Africa increased from about 12.57 million tons in 1990 to about 25.18 millions tons in 2008. This paper attempts to identify the factors which infl uence the eff ectiveness of production, consumption and foreign trade of milk in the Middle East and North Africa. The most important factors aff ecting consumption is the population, per capita income and produced quantity where a 1% increase in all of them results in increasing the quantity consumed by 1.3%, 2.86% and 0.611%, respectively. Milk sector provides employment opportunities for more than 25% of the working force in some Middle East and North Africa countries

    Price Volatility Modelling – Wheat: GARCH Model Application

    This paper is focused on the modelling of volatility in the agricultural commodity market, specifically on wheat. The aim of this study is to develop an applicable and relevant model of conditional heteroscedasticity from the GARCH family for wheat futures prices. The GARCH (1,1) model has the ability to capture the main characteristics of the commodity market, specifically leptokurtic distribution and volatility clustering. The results show that the forecasted volatility of wheat has a tendency towards standard error reversion in the long-run and the position of price distribution is closed to the normal distribution. The wheat production can be hedged against the price variability with long-term contracts. The price of wheat was influenced during the years of 2005 to 2015 by different events, in particular; financial crisis, increasing grain demand and cross-sectional price variability. The results suggest that agricultural producers should focus on short-term structural events the wheat market, rather than long-term variability

    Trade Impacts of Selected Free Trade Agreements on Agriculture: The Case of Selected North African Countries

    The objective of the study is to examine the impact of free trade agreements (FTA) with agricultural trade flow in general and dairy, vegetable, live animals, meat and sugar in particular. To achieve the objective the paper employs gravity model through compiling panel data. The study focuses on selected North African countries (Algeria, Egypt, Morocco and Tunisia) as reporting countries and the rest of the world as partner countries. Accordingly, the study finds that being a member of trade agreement (FTA) is positively associated with aggregate agricultural trade flow. In fact, trade agreement could increase agricultural trade flow by around 39 percent in trade volume (USD). Further, the study finds the potential of trade creation. In fact, the trade agreement with EU created a market for former Soviet countries (Latvia and Lithuania). Notably, due to the trade accord, the countries start exporting commodities such dairy and vegetable products. However, despite the results, the disaggregate agriculture fails to have a similar association. For instance, vegetable trade flow is positively influenced by FTA while live animals trade is affected negatively by FTA. Therefore, it requires vigilance when making a conclusion regarding the effect of FTA on disaggregates agriculture trade flow
